API 365 LC/MS/MS System
General Description
The
API 365 LC/MS/MS System is a triple quadruple mass spectrometer,
interfaced with a Shimadzu HPLC system. The ion source can be
Atmospheric Pressure (AP) Electrospray Ionization (usually applicable
for charged molecules such as proteins, peptides, and alkaloids)
or AP Chemical Ionization (usually applicable to non-charged molecules
such as lipids and terpenes).
It
can be used in LC/MS and LC/MS/MS modes. It can also be used in
MS or MS/MS moddes without HPLC by sample infusion.
Sample
Requirements:
For M. W. determination samples must be dissolved in solutions in
volatile solvents with concentrations >1uM (usually 5-50ul
volumes). Samples must be free of detergents, involatile salts,
and other modifiers. Sample solutions must be filtered with 0.2
micron filters. For peptides and proteins, best recommended solvent
is 0.1-0.2% aqueous formic acid/acetonitrile in the ratio: 6:4 (v:v).
