Live imaging directly involves inevitable tradeoffs of spatial resolution, temporal resolution and phototoxicity. This is especially true when interrogating imaging in three dimensions and/or three-dimensional samples over time (3D and/or 4D imaging) to obtain a complete picture of many dynamic subcellular processes.
Airyscan is a hexogen array of 32 concentrically arranged GaAsP detector; each detector approx. 0.2AU in diameter thus total detection area approx. 1.25AU to acquire most of the Airy disk all at once. The confocal pinhole itself remains open and does not block light, thus more photons are collected for the improved resolution and S/N.
